Training at VHIO and ITQB for proteomic analyses
Isabel worked with Francesc Canals, the responsible of the VHIO Proteomics Laboratory to identify Ehrlichia ruminantium (ER) proteins, focusing on post traductional protein modifications phosphorylation and glycosylation. The phosphorylation is related to key biological mechanisms such as bacterial division, metabolism and virulence and the glycosylation, very important to processes such as cell signaling, bacterial virulence and even pathogen evasion from immune system.
In the ITQB institute, Isabel worked in collaboration with Ana Varela Coelho, the responsible of the “Proteomics of Non-Model Organisms Laboratory” on the identification of proteins differentially expressed by host endothelial cells during ER replication. Important input of expertise from our project partner ITQB was performed. Various methods and tools have been mobilized through the ITQB proteomic platform (Decyder Software (GE Healthcare), in-gel digestion protocol with trypsin, MALDI-TOF-TOF mass spectrometer). Even if the results are still analyzed, Dr. Isabel reports “certain host cell proteins associated to the control of apoptosis and to cytoskeleton reorganization are highly impacted by ER at early stages of infection”.
